1252 Interferon mediated resistance of melanoma cells to oncolytic viruses is regulated by the melanocyte lineage transcription factor MITF

Oncolytic virotherapy represents a new approach in the treatment of metastatic melanoma. The efficacy this therapeutic is limited by release type I IFNs tumor microenvironment that can induce an antiviral state cells. Here we investigated ability to impair oncolytic virolysis panel human melanoma cell lines. We found cells with dedifferentiated phenotype showed increased IFN mediated resistance against viral oncolysis. Interestingly, IFN-responsive constitutively expressed interferon-related DNA damage signature (IRDS) was previously described resisted radio- and chemo-therapy. Using doxycycline-inducible gene expression system observed induction melanocyte lineage transcription factor MITF decreased IFN-mediated Transcriptome analyses revealed experimental activation modulates IFN-induced transcriptional network leading IRDS genes. Taken together our results imply acquisition MITF-low limits through IFN-responsiveness.
